Mitochondria use actin filaments as rails for fast translocation in Arabidopsis and tobacco cells

Fluorescently labeled proteins in transgenic Arabidopsis thaliana showed that mitochondria and peroxisomes moved at similar velocities. Movement of mitochondria was stopped by an actin-depolymerizing drug but not by a microtubule-depolymerizing drug. In living Arabidopsis cells and elongated cultured tobacco cells, mitochondria move along actin filaments as rails. The movement was not related to arrays of microtubules.

In variable incidence angle fluorescence microscopy, GFP-labeled Arabidopsis dynamin-related proteins (DRP1A and DRP2B), and clathrin light chain were observed as 200-500nm dot-like puncta around the plasma membrane in tobacco suspension cells. Moreover, these puncta moved to and away from the cell surface or also assembled and disassembled. These results raise the possibility that these three proteins are involved in membrane trafficking around the plasma membrane, including endocytosis.

We constructed a new T-DNA vector carrying the GUS reporter gene for gene trap analysis and the Ds element for targeted mutagenesis, and subsequently produced 51,000 gene trap lines. We analyzed the expression patterns of the GUS reporter gene in 20,000 gene trap lines. One hundred and forty integration sites of the T-DNAs in the gene trap lines were mapped on the genome. The T-DNA contains the Ds element therefore these lines can be used as donor loci for transposition.

The present investigation was undertaken to study the genetic variation within the species/varieties of Polyscias and Schefflera through RAPD and ISSR markers. A total of 164 and 69 distinct DNA fragments ranging from 300 to 2500 bp were amplified by using selected random RAPD and ISSR primers respectively. The correlation matrix indicate that there was significant correlation between ISSR and RAPD markers. This study will helpful for breeding programs as well as a major input into conservation biology of foliage crop.

The frequency of shoot regeneration from callus, node and shoot tip explants were higher on medium containing combinations of auxin and cytokinin than on medium containing cytokinin alone. Flowering can be induced in vitro in excised shoot cultures of Pentanema indicum. Adenine sulfate which has been found to be effective for shoot bud formation in P. indicum also supported the development of floral buds.

Agrobacterium-mediated transformation was performed in Japanese soybean variety, Kariyutaka. Because transgenic shoots of Kariyutaka formed in vitro precocious flower buds, T1 seeds were obtained in only six monthes after co-cultivation with Agrobacterium. These facts indicate that this transformation system can be used for the rapid development of transgenic soybean lines.
